I have a rectangle shape and inside it I have a circle path, how would I delete that path in the rectangle to make a transparent circle inside it?

You want to make a Compound Path. Select both paths, go to Menu>Object>Compound Path>Make [Command-8]. The circle will now appear as a hole in the rectangle.

I also find the Pathfinder Pallette To be most helpful. Make sure the Circle is ablove the rectangle in the layer stack, select both and click the Subtract from Shape Area icon.

I wonder if Ninjax meant to create the circle path as a cutout - not the whole circle?
In which case make the circle (no fill and light stroke), find and click the scale tool and scale down by a couple % or so and copy. Then make a compound path of these two circles. Now you can place an object underneath and use the Pathfinder (with both objects selected) to create the circle circumference as a cut out 'hole"
If that is so you get to use both techniques described by the other authors above. Nice!
